À compléter Janvier + Février Mars Mais Second dataset collection In 2009, a new set of apple leaves bearing pseudothecia of Venturia inaequalis were collected 1 to 3 weeks before bud break, from 0 to 10 days of bud break, and again 1 to 2 weeks after bud break in 8 countries. Leaf samples were immediately frozen at -18 ºC and sent to Québec and remained frozen until processsed. Triplicate leaf samples per site and collection date were set in plastic wire screens, designed to fit custom-built ascospore harvesting tower spore traps similar to previous de
signs (Gilpatrick et al. 1972). Twice weekly, the screens were placed facing downward over the trap on the large section of funnels connected to a suction device. The room temperature airflow in the traps was regulated with a plenum and optimized in such a way that spores naturally ejected from the wet leaves were impacted in a dry eppendorf vial set directly under each funnel. Samples were rewetted every twenty minutes for each 2 hour collection. Spores were resuspended in 200μl of water and their concentration was evaluated with a hematocytometer. Between ejections, samples were incubated moist at 18ºC to allow ascospore maturation but prevent discharge. Again, The maturation and discharge process was repeated until the ascospore supply was exhausted.
